Leech Hunter is a mini-game exclusive to Resident Evil 0 that I can say rather confidently that no one likes. Unfortunately, getting an A rank in this mode (collecting all 100 leech charms) not only nets you some achievements, but infinite ammo in all weapons on top of some exclusive weapons like the magnum revolver. You can play Leech Hunter by loading your save from your cleared file and selecting "Extra Game".
First off, in order to follow this guide properly, you need to get the correct placement of leeches and weapons. Yes, you can follow this strategy no matter what combination you get because the enemies will always be the same, but you will get worse weapons and different leech placement. So, the lucky combo is getting a blue and green herb at the foot of the stairs, no leech on the balcony. It doesn't matter if the leech next to the hatch is green or blue, because the same number of leeches will be in the same rooms, the colors will just be swapped.

Now that you have the right scenario, onto strategy. You need to collect 100 leeches, 50 of each color. Only Rebecca can pick up green charms and only Billy can retrieve the blue ones. Leeches CAN NOT BE DROPPED, so if you pick up just one leech that's an inventory slot rendered useless. Since you can only carry 10 leeches per item slot, it adds up quickly. So, the strategy here is to navigate the facility, kill all enemies, collect 90 leeches (50 Rebecca, 40 Billy), kill all respawned enemies, collect the 10 remaining leeches, and exit the facility. There's no saving, so make sure you have 2 hours free before attempting this.
In the first room, go down the ladder on the left. Move through the sewer and pick up the shotgun and shells as Billy, making sure Rebecca has her handgun equipped, and go through the door here. Immediately, you'll be ambushed by a group of 5 Hunters. Try to conserve ammo to take them down. When you see one jump at you, shoot it down for:
- Alley-oop!
Shoot down a Hunter or an Eliminator while they are jump attacking in the air.

With them all dead, use the herb if need be. If not, take it as Rebecca and pick up the fire key from the back of the room. Go back to the main hall and take the dining room door, dropping off the herb and your first aid sprays here for now. In the dining room, use your pistols to kill all the zombies. Unlock the kitchen door with the fire key, then grab the knife from the cart next to the door as Billy. Go through to the hallway that leads to the basem*nt now. Knife the 2 zombies lying prone on the floor, then drop the knife here. Go down the stairs to the animal statue room, telling Rebecca to wait at the door and equip your shotgun. Go over to the area where the gate was raised in the campaign, and slowly back into the area. When you hear the Hunter, run out and turn around, blasting it. With that Hunter dead, go over to where you saved Rebecca from falling in the campaign. Before rounding the corner, do the same backing in technique to lure this Hunter out for a kill as well. With both of them dead, regroup and get out your pistols. Then go through the right side door that was in the opened gate area by the puzzle.
The zombies in here are armored, so they may take more shots. Collect the grenade launcher on the bed as Rebecca, as well as any herbs you need before leaving. Go down the door at the end of the hall. There's a few zombies in here, as well as one right by the door that will stand up immediately. Focus fire on him, at least knocking him back to deal with the other zombies. Once this room is clear, go into the room you saved Rebecca in the campaign. This is a tight room now infested with zombies, so hopefully you can keep them knocked down long enough to avoid damage. With them dead, go down to the stairs here. Make Billy wait at the top as Rebecca goes through with the grenade launcher, using 1 shot each on every spider here. Once they're dead, switch to the pistol and shotgun to tackle the room on the right.
This one has 2 Plague Crawlers, so use your weapons to keep them suppressed before killing them. Collect the herb here and move back all the way to the main hall, passing through the main areas to reach it. Drop your healing items in the main hall health cache you're building before making Billy wait and sending Rebecca through the western hall alone. Equip your grenades and go into the bathroom. Once inside, target the Mimic. As soon as you see/hear it change, shoot it until it does its little death dance, earning you:

Defeat Mimicry Marcus with a Molotov co*cktail or a Napalm Grenade.
Pick up the shotgun and handgun ammo from inside this room, then return to the hall. Continue down the hall for another Mimic. Get behind it and wait for the change before putting your flame rounds into it for another painless kill. You can take the 3 herbs from the projector room if you wish before returning to the main hall. Go into the middle section (where the Marcus painting was) and use your flame rounds on the spiders in this hall. Move Billy in here, give him the shotgun shells, equip your pistols, and go through the right side door. This room has a zombie cluster you should be able to keep at bay damage free.
Once they're taken care of, equip the shotgun and go into the torture room. There's another group of Hunters here, so be sure to shoot them down quickly. There's an herb on the nearby chair if need be. If you finish the fight and have 1-2 shells left in your shotgun, don't reload it. Just drop it here, pick up the shotgun next to the iron maiden, grab the shells, and leave this room. Run down the hall to what used to be the safe room. As soon as you get inside, 2 Eliminators will jump you. Keep the fire on them with the shotgun until they're dead; they take a ridiculous amount of damage. Once these 2 are dead, go through the door. 2 more Eliminators are in here, but will usually just keep rushing you at the door until they're dead. Once they are, take the magnum and ammo in here as Billy before returning to the main hall.
Alright, that's the basem*nt and first floor relatively cleared out. Go back to the dining room, to the basem*nt hall, and through the door on the right with the magnum equipped. This room has 4 Hunters that will try to pounce immediately, but 1 magnum shot apiece will kill them without worry. With the Hunters dead, switch back to the pistols and climb the ladder. Clear out the balcony of zombies here, which is an easy task. With them gone, unlock the door at the end and pass through. Take your magnum back out and go through the knight door, right into the arms of more Hunters. Kill them with a shot, then take out your pistol and go into the medical room at the end of the hall. The zombies can close in on you here, so if you feel a shotgun blast is necessary, use it by all means. With the zombies dead, drop Billy's pistol here in favor of the custom handgun on the bed, plus the first aid spray if it's needed. Return to the conference room.
Go through the other door here with the magnum equipped, ready to deal with 3 more Hunters, leaving you with 2-3 magnum shots. Get out your pistols and go into the library, dropping the 2 zombies in here. Now head through the side library door alone, as going with both players means whoever is in front gets munched on automatically. Kill the 2 standing and 1 prone zombies in here, hop up on the small ledge on the right, grab the handgun ammo from the wall, and leave the library. Go down the hall and use the fire key here, throwing it away after. You have a choice in this room, and can either shoot all 4 prone zombies, get munched on and kick their heads in, or go back to the typewriter in the conference room and grab the knife there to stab them all to death. Whatever situation works best for you. One of the zombies is hidden under the pushable table, so watch out for that one and make sure 4 are dead before returning to the main hall.
Go through the door on the left here to kill some crows, not a big deal at all. There's a gun in here with 15 shots, remember it in case you need it later. Leave this room and head across the balcony to the room you got Microfilm B in the campaign, using combined pistol power to kill all the zombies in here. With them dropped, go through the door to the left of this one and through the door on the left in this hall. Take out the Crawler's in here with shotgun shells, before arming Rebecca with the grenades and sending her into the next room alone. In this hall, immediately go left into the broadcasting room. Wait for the transformation from the Mimic before taking him down with the fire rounds. Exit this room and deal with the Mimic in this hallway the same way. Now go into the next hall, and kill the final Mimic in here with the rounds. Switch back to Billy.
Go through the blue door and into the hallway, going into the piano room. Use your knife to kill the prone zombies in here (one generously drops a green+red herb mixture if you're hurting) and drop the knife when they're dead. Leave here, go into the hallway, and re-link with Rebecca, both of you with pistols. Go into the room all the way to the right of your characters, killing the cluster of zombies (including the crawler) in this room. Have Billy equip his magnum and go through the door at the other end of this hallway, dropping the 2 Hunters in here with a shot each. Grab the grenade launcher under the table in the small room as Rebecca. Now return to the Centurion room with Rebecca. Go upstairs and use the remaining flame rounds you have on the Plague Crawlers here, as well as the single shot grenade and any shotgun ammo you have. Drop both grenade launchers here and go through the knight door. Pistol the zombie on the walkway, then go over to the control area and aim down to kill the remaining few. That's the entire main batch of enemies dead, so give Billy all the handgun ammo and have him drop the shotgun, magnum, and anything that's not the custom handgun and ammo for it in this room. Give Rebecca only a fully loaded handgun. Let the hunt begin!
Note: I'll just be naming off every room in sequence with the number of leeches in each. Count them as you pick them up to make sure you're not stuck wandering around at 99 leeches

Note: Now you should have 90 leeches, 40 for Billy and 50 for Rebecca. Before getting the last 10 leeches for Billy, go to the following locations and kill the respawned enemies.
Waterfall room, animal statue room, balcony, knight door hall, medical room hall, basem*nt room next to the torture room, kitchen. After killing the last enemies, you can drop your ammo and unlock:

Defeat all enemies in Leech Hunter.

After collecting your 100th leech, return to the main hall. Exit through the knight doors you went through to get the tablet using the battery in the campaign. The game will tally up all of your leeches, giving you access to unlimited ammo for everything, some new weapons, and:

With those out of the way, start another Leech Hunter game. This game, give all of Rebeccas ammo to Billy. Have Rebecca follow you around the whole time, collecting ONLY HER GREEN LEECHES. The route and strategy remain the same, only that Rebecca is picking up leeches and dropping guns when she runs out of room. Once all 50 of her leeches have been collected, return to the main hall and exit the area for:
- Who Needs Partners?
Collect all of only one color of Leech Charms.

Now, start up Leech Hunter one final time. Keep restarting until you get 2 green herbs in front of the stairs. Pick them up and combine them, then give the mixture and your first aid spray to Billy. Separate from Rebecca, go to the dining room, and grab the knife directly next to you. Go back across the main hall to the hallway here. Equip the knife, and start hacking away at the Mimic in this corridor. It may take all the health items you have plus an herb from the projector room next to you, but once the knife slays the Mimic you'll unlock:
